Alkaline Protease

The alkaline protease is prepared by deep liquid fermentation of Bacillus licheniformis. It can hydrolyze macromolecular protein to produce polypeptides or free amino acids.

Features:

1. Appearance: light brownish powder with less than 8% of water content.

2. Activity: 200,000 U/g

3. Temperature: 40-50°C

4. pH: 7.0-12

Definition of the unit of the enzyme activity: One unit (U/g) is defined as the amount of enzyme needed to hydrolyze casein to produce 1 μg of tyrosine measured as the absorbance value in furin reagent chromogenic reaction in sodium borate - sodium hydroxide buffer (pH 10.5) at 40°C in 1 min (total reaction time is 10 min) (QB/T1803-1993).

Applications:

1. The alkaline protease is a non-toxic protein with no side effects, which belongs to the serine endopeptidase. In food industry, it can be used to hydrolyze protein to produce peptides or amino acids which can form protein lysate with a unique flavor.

2. The alkaline protease can be added in ordinary and concentrated detergent powder and liquid detergent, which can effectively remove blood stains, eggs, dairy products, or gravy, juice and other protein stains. It also can be used as medical washing reagent to clean biochemical instruments.

3. The alkaline protease can be used for silk degumming: pretreatment – dehydration – treatment with acid – (treatment with protease at 18-24U/ml, 0.75% of soap at 48-50 ⁰C for 30-60 min, PH 7-8) – dehydration – peeling off the skin – rinse – dehydration – fluffing - drying - fluffing.

4. The alkaline protease can also be used for leather depilation: raw leather - washing – degreasing – alkaline expansion – dehairing – splitting – dealkalization – enzymatic dehairing – washing - pickling – tanning.

Storage:

Store in dry and cool (below 25°C) area with ventilation. During transportation, avoid direct exposure to strong sunlight, rain and moisture. The enzymes will lose activity at high temperature or under strong sunlight. The enzyme activity can be maintained at least 85% when stored for 6 months. The alkaline protease can be stored for long period at 5°C. It is corrosive to skin and direct contacting with skin and eyes should be avoided.
