Cultivating Bacillus subtilis
The cultivation conditions of Bacillus subtilis are very specific and strict. When cultivating, one should be cautious and follow the directions carefully. These conditions mainly include the following aspects.
Composition and Ingredients:
The culture medium for Bacillus subtilis usually contains 10g of peptone, 5g of yeast powder, 10g of Sodium Chloride, and 1.0L of distilled water, with a pH value of 7.0. Solid culture medium also requires the addition of 1.5% -2% agar powder.
Aeration:
Bacillus subtilis is an aerobic bacterium that needs to be cultured in an aerobic environment.
Cultivation Temperature:
Usually cultured at 37 ℃, the conventional cultivation time for Bacillus subtilis and most bacteria is 24-48 hours.
Storage Conditions:
Glycerol strains should be stored at -80 ℃, while agar strains can be stored at 4 ℃.
Caution:
When cultivating Bacillus subtilis for the first time, activate according to recommended conditions and avoid using other types of culture media or conditions to prevent bacterial inactivation. Bacterial waste should be disposed of after high-pressure sterilization treatment.
